Myxengo's proprietary human naive scFv Phage Library is based on a combinatorial library of human VH and VL (kappa and lambda) fused to a minor coat protein (pIII) of M13 phage. The displayed scFv is expressed at the N-terminus of pIII. The peptide is followed by a short spacer (3X Gly-Gly-Gly-Ser linker) and then the wild-type pIII sequence. The library consists of more than 5X10E10 electroporated scFv sequences amplified from naive human B cells. we use approximately 100 copies of each sequence for a target. We separately maintain the kappa library and lambda library in order to avoid bias toward to a class of light chain for a target. 95% of Myxengo's human naive scFv Phage Library contains function scFv genes without stop codon or frame shift in the gene. Furthermore myxengo developed a new selection method to sort out non-functional scFv from the selection pool resulting in highly efficient selection and highly diverse leads with better affinity. |
